(1) 天然活性成分之免疫調節機轉及功效評估
薏仁是具有免疫調節功能的機能性穀類食品,為探討薏仁改善過敏性疾病之潛力,首先利用細胞實驗初步篩選薏苡種皮及薏仁麩皮抑制嗜鹼性細胞(RBL-2H3)去顆粒化之活性,結果發現薏仁具有抑制嗜鹼性細胞釋放組織胺之能力,因此推測薏仁具有改善過敏發炎反應之功效;相關研究成果發表於J. Agric. Food Chem. (2011)J. Ethnopharmacol. (2012)。進一步確認薏仁對活體免疫調節的功效,在小鼠過敏模式(OVA)下,薏仁麩皮萃取物可顯著降低過敏動物體內特異性IgE抗體生成,並調節Th1/Th2免疫反應來減緩動物體內過敏症狀;此研究成果發表於Food Chem.Toxicol. (2012)

(2) 食材中活性成分之分離與鑑定
活性成分是科學界所重視的重要指標,因此實驗室將針對具活性之食材進行活性成分的分離純化與鑑定。過去的研究成果包括:薏仁麩皮中活性成分之分離,結果分得22個化合物,其中15個為薏仁中首次分離,包括1個天然物首次發現之新穎化合物auronol以及11個類黃酮、2個色酮和1個酚酸酯;相關研究成果發表於Food Chem. (2011),並將新化合物的抗發炎功效申請美國專利(US 20130158107 A1)。金針花一系列抗氧化天然物之分離,此成果發表於J. Agric. Food Chem. (2011)黃芩純化物減緩肝纖維化之功效,證實baicalein可有效抑制肝臟內皮細胞的活化,並阻止肝臟星狀細胞纖維化,研究成果已發表於J. Ethnopharmacol. (2013)。而在TNF-α引起的肝發炎細胞模式中,甘草中的glycyrrhetinic acid可藉由抑制NF-κB活化來達到減緩慢性肝病形成,研究成果已刊載於J. Agric. Food Chem.(2014)

(3) 食品品質管制分析平台之建立
面對食材或健康食品等複雜之化學組成,建立品質管制之平台是必須努力的。因此茲將薏仁分離出之類黃酮類成分,以HPLC/MS利用m/z的比對進行定性分析,可做為日後薏仁麩皮產品品質管制之分析平台,此一方法已發表於Food Chem. (2011)。博士後期間協助桃改場完成栽培天麻活性成分分析及HPLC/UV檢測平台。目前為建立中華藥典資料庫,利用液相層析搭配四極柱-飛行時間串聯式質譜(UPLC-ESI-Q-TOF/MS)建立中草藥植物化學成分之指紋圖譜,並以主成分分析做為中草藥品質標準化之分析平台。

(4) 利用核磁共振及質譜分析進行代謝質體學之研究
中草藥研究常常遇到複方藥的複雜性,然而在臨床使用上並非將單味藥的單一活性成分做為治療處方,因此,如何探討中草藥這種多靶點、多成分的治療功效,是需要國內研究專家學者一起努力的。實驗室利用代謝質體學的觀點再搭配系統生物學的分析,探討非破壞性之核磁共振光譜究應用於傳統中藥方劑對於缺血性中風小鼠之療效,研究結果顯示中風小鼠因腦部缺血造成厭氧代謝使乳酸(lactate)及酮體(ketone body)大量產生,鈣離子大量流入(Ca2+ influx)使細胞膜崩解而釋出興奮性神經毒,而在給予【補陽還五湯】後對於缺血性中風小鼠具有保護延長壽命之作用;此研究結果提供【補陽還五湯】對栓塞型中風療效的有力科學證據,並且提供評估中藥方劑療效的簡易實驗平台,研究成果發表於Metabolomics (2012)。爾後更進一步利用UPLC-ESI-Q-TOF/MS進行iTRAQ-base proteomics (蛋白質體學)的研究,相關研究發表於PLoS One (2015)

(5) 以代謝質體學進行中醫體質科學化之人體試驗
實驗室與國家中醫藥研究所林雲蓮研究員,協同台北榮總傳統醫學科陳方佩主任及同欣堂中醫師林以正合作,執行人體試驗收集受試者血液後,分別利用NMR及UPLC-ESI-Q-TOF/MS等方法,以代謝質體學的角度切入中醫脈診的研究,期望能以科學化的方法解釋中醫脈診的理論;相關成果發表於Evid-based Compl. Alt. (2016)
