            多醣与淀粉化学及机能性醣类食品的开发,为本研究室的主要研究兴趣,目前的研究重点是利用富含多醣之菇菌类(如香菇、灵芝、巴西洋菇等)与中草药植物(如金线莲、黄耆、黑豆等)为主要材料,来开发机能性醣类保健食品;另外,利用物理与化学的分析方法,探讨富含淀粉与多醣胶质,且已具工业化生产之谷类、豆类、根茎类作物及海藻等原料的物理与化学特性,再以物理、化学或酵素的方法加以修饰,以提高其在食品加工过程与产品中的功能性与安定性。本研究室与国内多个研究单位有合作关系,是「灵芝研究联盟」的成员,也参与行政院农委会农业试验所「化学诱变米种选育研究」的工作,近期研究的成果与方向有:
 
机能性醣类方面:
  1. 利用高效能分子筛层析法配合多角度雷射光散射技术,分析灵芝(Ganoderma lucidium)热水萃取液中多醣之分子量分,并以刺激人类周边血液单核细胞产生肿瘤致死因子(TNF-α)作为活性指标,结果显示(1®3)-b-D-glucans 为灵芝子实体中的主要活性多醣,其含量佔总中性多醣的31%,其分子量为2000 kD,其分支成度为0.35。
  2. 利用(1®3)-b-D-glucans与萤光染剂的选择性结合,建立一个简易的萤光呈色法,用以分析菇菌类产品中(1®3)-b-D-glucans的含量。
  3. 利用系列酵素反应并配合高效能阴离子层析法,建立一个可定量分析菇菌类产品中(1®3)-b-D-glucans的含量,并可同步分析其分支度的方法。
  4. 利用上述分析方法为品质评估指标,评估栽培方法对灵芝(1®3)-b-D-glucans的生成量、分子量与分支度的影响,发现菌丝体培养所生成的指标性多醣,在含量、分子量与分支度等,均有显着的影响,正配合「跨部会健食品研发计画」的其他子计画,寻找可生产更高活性多醣的制程。
  5. 探讨台湾金线莲 (Anoectochilus formosanus Hayata) 水溶性多醣的分子特征,发现含有近似比率的中性与酸性多醣,且此两种多醣对人类周边血液单核细胞,均有刺激产生肿瘤致死因子(TNF-α)与间白素-6(IL-6)的活性。
  6. 探讨山药水溶性多醣之免疫活性,发现对J774.1类老鼠巨噬细胞株有抑制活性,对人类周边血液单核细胞有刺激产生肿瘤致死因子(TNF-α)与间白素-6(IL-6)的活性。
 
多醣与淀粉分析与修饰:
  1. 利用细菌淀粉液化酵素部分降解生淀粉颗粒为工具,研究淀粉颗粒的降解型态与淀粉颗粒组成构造的相关性与颗粒中直链淀粉与磷脂质错合物的分布,并利用此方法产生具有多孔状的颗粒,目前正继续探讨这种多孔状淀粉的特性与应用性。
  2. 利用酸水解淀粉颗粒所产生之短链淀粉分子探讨分子的重排,成功利用乙醇浓度控制产生具有x-ray绕射图为A-、B-与混合型的C-型结晶。
  3. 探讨化学诱变稻米系列的淀粉性质,初步分析发现由同源米种所诱变产生的稻米系列,其直链淀粉含量的分布非常广泛,其范围可涵盖糯性至28%,此系列之稻米具有非常相近的基因背景,是研究基因、合成酵素与淀粉结构相关性的良好材料。
  4. 探讨不同干燥方法,对山药粉末复水泥之黏稠与成胶性质的影响,并以松饼小西饼、面条与沙拉酱等加工型式,探讨山药的烘焙与加工性质。
 
醣苷分析:
  1. 探讨台湾产山药(Dioscorea spp.)中固醇类皂素的含量与型态,发现台湾产山药块茎只有少数的栽培种含有diosgenin(薯蓣皂配基),且均以醣苷的型态存在,在每克块茎干重中diosgenin的含量范围为41~68 μg。为分析此类醣苷发展出一个新的高效能层析法,可同步分析furostanol与spirostanol glycosides 两群不同皂素,同时也鑑定出两个首次在山药中发现带有四个醣基的皂素。(与孙璐西教授合作计画)
  2. 探讨台湾产山药植物固醇含量与型态,发现块茎中含量在栽培种间差异很大,但均以自由态与醣苷化形式存在,未发现有可酯化态植物固醇,同时也发展出一个气体层析法,可同步分析植物固醇与薯蓣皂配基。
